Sirtuins (SIRT1–SIRT7) are NAD+-dependent protein deacetylases. SIRT1 (nuclear) and SIRT3 (mitochondrial) are the most studied: - SIRT1 deacetylates FOXO, p53, and PGC-1α, influencing stress resistance and mitochondrial biogenesis - SIRT3 deacetylates mitochondrial proteins, regulating oxidative metabolism and ROS detoxification NAD+ is the obligate coenzyme — without sufficient NAD+, sirtuin activity collapses. This makes NAD+ supplementation a standard control in sirtuin-focused in-vitro assays.
